结直肠癌组织中脂类伸缩振动的拉曼光谱

为了探索脂类分子结构的变化与细胞癌变的关系,应用共聚焦显微拉曼光谱系统,检测结直肠癌组织及邻近正常组织的拉曼光谱,得到归属于脂类CH_2对称伸缩振动在2857 cm~(-1)和反对称伸缩振动在2891 cm~(-1)的拉曼峰以及归属于脂类和蛋白C—H伸缩振动在2938 cm~(-1)的拉曼峰。实验结果显示,结直肠癌组织和邻近正常组织的拉曼光谱存在以下显著的差异:1)与正常组织相比,结直肠癌组织中位于2857 cm~(-1)和2891 cm~(-1)的拉曼峰强度相对较弱,而2938 cm~(-1)拉曼峰的强度相对较强。2)结直肠癌组织中在2891 cm~(-1)和2857cm~(-1)的拉曼峰强度比值I_(2891)/I_(2857)小于正常组织的比值。用SPSS16.0统计学软件中平均值之间的比较功能计算出正常组织的I_(2891)/I_(2857)平均值为2.21,癌变组织的I_(2891)/I_(2857)平均值为1.69。研究结果表明,当正常细胞转变为癌细胞时,脂类分子的侧向堆积程度下降,细胞膜流动性增加,脂类与蛋白的相互作用增强,揭示了脂类分子伸缩振动的拉曼光谱与细胞癌变的关系。 In order to explore the relationship between changes of lipid molecular structure and canceration of cells, the Raman spectrum of colorectal cancer tissues and adjacent normal tissues was detected by confocal Raman microscopy. The symmetry stretching vibration of CH 2 at 2857 cm ~ (-1) and Raman peaks of antisymmetric stretching vibration at 2891 cm ~ (-1) and Raman peaks at 2938 cm ~ (-1) which belonged to stretching vibration of lipids and proteins at C-H. The results showed that the Raman spectra of colorectal cancer tissues and adjacent normal tissues showed the following significant differences: 1) Compared with normal tissues, the Raman spectra of 2857 cm ~ (-1) and 2891 cm ~ (-1) ) Raman peak intensity is relatively weak, while the 2938 cm ~ (-1) Raman peak intensity is relatively strong. 2) The intensity ratio of I (2891) / I (2857) at 2891 cm ~ (-1) and 2857 cm ~ (-1) in colorectal cancer tissues was smaller than that of normal tissues. The average value of I_ (2891) / I_ (2857) in normal tissues was calculated to be 2.21 by using the comparison function between the means in SPSS16.0 statistical software, and the average of I_ (2891) / I_ (2857) in cancerous tissues was 1.69 . The results show that when the normal cells into cancer cells, the lateral accumulation of lipid molecules decreased, the membrane fluidity increased, lipid and protein interactions enhanced, revealing the stretching vibration of lipid molecules Raman spectra and cells The relationship between cancer.
