目的 :通过探讨不同给苯剂量、苯作用时间的小鼠反应 ,筛选建立苯诱发小鼠再生障碍性贫血模型的方法。方法 :予雄性CD1小鼠每周三次皮下注射“苯 +玉米油”混合液 ,按给苯剂量不同分为四组 :B1组(0 .5ml/kg)、B2组 (1.0ml/kg)、B3组 (1.5ml/kg)、B4组 (2 .0ml/kg) ,以单纯注射玉米油组 (A组 )为对照 ,观察小鼠一般状况、血象、骨髓象、骨髓有核细胞计数 ,并进行肝、脾和胫骨的组织病理学观察。结果 :与对照组相比 ,B4组在给苯 2 5次后体重和脾脏指数分别下降 18.5 1%和 6 3.86 % (P <0 .0 1) ,白细胞、红细胞、血红蛋白、血小板、网织红细胞绝对值和骨髓有核细胞计数分别下降 74 .97% (P <0 .0 1)、4 1.6 6 % (P <0 .0 1)、30 .96 % (P <0 .0 1)、2 4 .77% (P <0 .0 5 )、4 1.4 7% (P <0 .0 5 )和 6 1.6 1% (P <0 .0 1) ,骨髓涂片检查和病理形态学观察均显示造血细胞减少、脂肪细胞和淋巴细胞等非造血细胞增加 ;综合以上结果提示本实验中苯诱发再障模型建立成功。结论 :予CD1小鼠每周三次皮下注射 2ml/kg体重的苯 ,累计给苯 2 5次 ,共 5 0ml/kg ,初步建立了苯诱发小鼠再生障碍性贫血模型的实验方案。
OBJECTIVE: To establish a model of benzene-induced aplastic anemia in mice by investigating the response of mice exposed to different doses of benzene and benzene. Conclusion: CD1 mice were injected subcutaneously with 2 ml / kg of body weight benzene three times per week for a total of 25 times with benzene for a total of 50 ml / kg. The experimental protocol of benzene-induced aplastic anemia in mice was initially established.
