目的 观察体外培养的正常猴眼小梁细胞及睫状肌细胞中组织金属蛋白酶抑制剂(tissueinhibitorsofmetalloproteinase ,TIMP)的表达 ,探讨生理状态下基质金属蛋白酶 (matrixmetalloproteinase,MMP)及TIMP在小梁网房水流出及葡萄膜 巩膜房水流出通道中的作用。方法 采用Reverse zymography技术 ,检测猴眼小梁细胞及睫状肌细胞中TIMP及MMP的表达。结果 猴眼小梁细胞及睫状肌细胞培养液中均可见TIMP 1、TIMP 2及TIMP 3表达 ,小梁细胞中MMP/TIMP比值明显高于睫状肌细胞。结论 正常状态下小梁细胞外基质的降解能力明显高于睫状肌细胞 ,可能是由于传统的小梁网房水流出通道作用强于葡萄膜 巩膜房水流出通道的作用。
Objective To investigate the expression of tissue inhibitor of metalloproteinase (TIMP) in cultured normal ocular trabecular cells and ciliary muscle cells in vitro and to explore the role of matrix metalloproteinase (MMP) and TIMP in trabecular meshwork Outflow and uveal scleral aqueous outflow channel role. Methods Reverse zymography was used to detect the expression of TIMP and MMP in monkey trabecular meshwork cells and ciliary muscle cells. Results The expression of TIMP-1, TIMP-2 and TIMP-3 in the trabecular meshwork cells and ciliary muscle cells in trabecular meshwork cells was significantly higher than that in ciliary muscle cells. CONCLUSION: Under normal conditions, the degradation capacity of trabecular meshwork extracellular matrix is significantly higher than that of ciliary muscle cells, which may be due to the effect of trabecular meshwork water outflow channel stronger than that of uveal aqueous scleral outlet channel.
