目的:检测伴有kazal域的富含半胱氨酸的逆转诱导蛋白(RECK)、晚期糖基化终末产物受体(RAGE)及基质金属蛋白酶9(MMP-9)在鼻咽癌(NPC)组织中的表达情况及其相关性。方法:免疫组织化学方法检测64例NPC(伴颈淋巴结转移者30例,无颈淋巴结转移者34例)和30例慢性鼻咽炎(CNT)组织中RECK、RAGE和MMP-9蛋白的表达。结果:RECK在CNT及NPC组织表达极低,但周边炎症细胞及癌巢周边基质中RECK强表达,在伴与不伴颈淋巴结转移组中的阳性率分别为3.3%(1/30)和11.8%(4/30),2组间差异无统计学意义(P>0.05)。RAGE表达阳性率在CNT和NPC组织分别为100%(30/30)和70.3%(45/64),MMP-9则分别为46.7%(14/30)和78.1%(50/64);RAGE在伴与不伴颈淋巴结转移组中的阳性率分别为86.7%(26/30)和55.9%(19/34),MMP-9则分别为90.0%(27/30)和67.7%(23/34),组间差异均有统计学意义(P<0.05);在NPC组织中,RECK与MMP-9的表达呈负相关(r=-0.369,P<0.01),RAGE与MMP-9呈正相关(r=0.471,P<0.01)。结论:在NPC组织中,RECK、RAGE较CNT组织表达下调,MMP-9上调;在伴转移的NPC组织,RECK表达下调,RAGE、MMP-9则上调。三者的异常表达可能与NPC的进展有关,RECK、RAGE可能通过调节MMP-9的表达参与NPC的侵袭转移过程。
OBJECTIVE: To detect the expression of RECK, RAGE and MMP-9 with kazal domain in nasopharyngeal carcinoma (NPC ) Tissue expression and its correlation. Methods: The expressions of RECK, RAGE and MMP-9 in 64 cases of NPC (30 cases with cervical lymph node metastasis, 34 cases without cervical lymph node metastasis) and 30 cases of chronic nasopharyngitis (CNT) tissues were detected by immunohistochemistry. RESULTS: The expression of RECK was very low in CNT and NPC tissues, but RECK was strongly expressed in peripheral inflammatory cells and surrounding cancerous nests. The positive rates of RECK were 3.3% (1/30) and 11.8% in patients with and without cervical lymph node metastasis % (4/30), no significant difference between the two groups (P> 0.05). The positive rate of RAGE expression was 100% (30/30) and 70.3% (45/64) in CNT and NPC tissues respectively, and 46.7% (14/30) and 78.1% (50/64) in MMP-9. The positive rates of MMP-9 in the group with and without cervical lymph node metastasis were 86.7% (26/30) and 55.9% (19/34), respectively. The positive rates of MMP-9 were 90.0% (27/30) and 67.7% (P <0.05). There was a negative correlation between the expression of RECK and MMP-9 in NPC (r = -0.369, P <0.01), but there was a positive correlation between RAGE and MMP-9 (r = 0.471, P <0.01). CONCLUSION: In NPC tissues, RECK and RAGE are down-regulated and MMP-9 is up-regulated compared with CNT. In NPC tissues with metastasis, RECK expression is down-regulated while RAGE and MMP-9 are up-regulated. The abnormal expression of the three may be related to the progress of NPC, RECK, RAGE may participate in the invasion and metastasis of NPC by regulating the expression of MMP-9.
