本文介绍了抗体研制的当代先进技术和今后的发展趋势。概述了采用体细胞遗传技术的杂交瘤单克隆抗体、双特异性功能抗体的研究,到采用基因重组技术研制鼠源性抗体的Fab或Fv小片段,再嫁接人源性FC片段组成功能完整的人化抗体,消除排斥反应以及采用位点突变和先进的选择方法提高研制抗体亲和力。作者预言通过技术创新和老技术改进两者的结合,人造抗体的发展将会为人类作出伟大的贡献。
This article introduces the contemporary advanced technology for antibody development and future trends. Summarizes the research of hybridoma monoclonal antibody and bispecific antibody using somatic cell genetic technology to develop a Fab or Fv small fragment of murine antibody using gene recombination technology and then grafts the human FC fragment with complete function Humanized antibodies, elimination of rejection, and site-directed mutagenesis and advanced selection methods improve antibody affinity. The authors predict that the development of artificial antibodies will make a great contribution to mankind through the combination of technological innovation and old technological improvements.
