α—FP 是临床诊断肝癌的一个重要指标。本文将醋酸纤维素-抗体-抗原膜与自制的氯电极研制成α—FP 非标记免疫传感器。研究了载体膜组成对电极灵敏度的影响;采用均匀设计、单纯形法优化了膜结合抗原抗体复合物的实验条件和参数;与同类电极相比,该电极使用简便,响应快(≤3min),灵敏度高,重视性和稳定性均好;在最佳测试条件下,电极线性范围为0-600ng/ml,检测下限26ng/ml,相关系数0.9925,测量600ng/mlα—FP 时的标准偏差为30ng/ml;人体血清中所含主要免疫球蛋白对α—FP 的测量均无干扰;在用于实际血清样品测量中取得满意结果。
α-FP is an important indicator of clinical diagnosis of liver cancer. In this paper, cellulose acetate - antibody - antigen membrane and self-made chlorine electrode developed α-FP non-labeled immunosensor. The influence of the composition of the carrier membrane on the sensitivity of the electrode was investigated. The experimental conditions and parameters of membrane-bound antigen-antibody complex were optimized by uniform design and simplex method. Compared with the same type of electrode, The sensitivity, importance and stability were all good. Under the optimal test conditions, the linear range of the electrode was 0-600ng / ml, the detection limit was 26ng / ml, the correlation coefficient was 0.9925, and the standard deviation of 600ng / mlα-FP was 30ng / ml; the main immunoglobulin contained in the human serum had no interference with the measurement of α-FP; satisfactory results were obtained in the actual serum sample measurement.
