目的:了解肝细胞生长因子(HGF)及其受体基因在人肝癌和癌旁肝组织中表达的变化规律,以及这种变化与肿瘤生物学行为的关系。方法:采用地高辛标记的HGF及其受体DNA探针,检测26例人原发性肝癌及其癌旁肝组织中HGF及其受体基因的表达。结果:HGF在人肝癌和癌旁肝组织中虽都有表达,但阳性率不及其受体基因表达,HGF受体在肝癌和癌旁肝组织中均呈过度表达,且肝癌组织中杂交信号明显高于癌旁肝组织,有肝内外转移者其表达更高。结论:HGF受体基因过度表达与肝癌的转移密切相关,HGF及其受体系统对调节肿瘤生长、转移起重要作用
OBJECTIVE: To investigate the changes of hepatocyte growth factor (HGF) and its receptor gene expression in human liver cancer and adjacent liver tissue, and the relationship between this change and tumor biological behavior. METHODS: The expression of HGF and its receptor genes in 26 cases of primary hepatocellular carcinoma and its adjacent liver tissues were detected by digoxigenin-labeled HGF and its receptor DNA probes. RESULTS: Although HGF was expressed in human hepatocellular carcinoma and para-carcinoma liver tissues, the positive rate was not associated with its receptor gene expression. HGF receptor was over-expressed in both hepatocellular carcinoma and para-carcinoma tissues, and the hybridization signal was apparent in hepatocellular carcinoma tissues. It is higher than that in adjacent liver tissues, and it is expressed more highly in livers with metastases. Conclusion: Overexpression of HGF receptor gene is closely related to the metastasis of HCC. HGF and its receptor system play an important role in the regulation of tumor growth and metastasis.
