虫媒黄病毒包括一大类对人类致病的病原体,基因组为单股正链RNA,依次编码3种结构蛋白和7种非结构蛋白。其中,非结构蛋白NS5具有RNA依赖性RNA聚合酶、甲基转移酶等多种酶活性,在病毒基因组的复制过程中发挥关键作用。最近的研究发现,NS5能够通过多种机制调控宿主的抗病毒反应。NS5的多功能特点使其成为潜在的抗病毒药物靶点并受到广泛关注。本文综述了虫媒黄病毒NS5蛋白生物学功能的主要进展。
The insect-borne flaviviruses include a large group of pathogens that cause disease in humans. The genome is single-stranded positive-stranded RNA, which in turn encodes three structural proteins and seven non-structural proteins. Among them, the non-structural protein NS5 has various enzyme activities such as RNA-dependent RNA polymerase and methyltransferase, and plays a key role in the viral genome replication. Recent studies have found that NS5 can regulate the host’s antiviral response through a variety of mechanisms. The versatile nature of NS5 makes it a potential target for antiviral drugs and is receiving widespread attention. This review summarizes the major advances in the biological functions of the NS5 protein of the flavivirus.
