目的探讨非洛地平逆转代谢综合征(MS)心肌纤维化的作用机制。方法雄性Wistar大鼠33只随机分为对照组(12只)和代谢综合征组(21只),采用10%高果糖饮水8个月建立MS动物模型,随机将9只代谢综合征组大鼠给予非洛地平。并采用苦味酸天狼猩红染色法检测大鼠左室心肌胶原含量,RT-PCR法检测心肌白细胞介素-18(IL-18)mRNA表达水平。结果与对照组相比,MS大鼠左室心肌组织胶原含量升高(P<0.01);IL-18mRNA表达水平升高(P<0.01);应用非洛地平干预治疗后,上述异常均减轻。结论非洛地平通过抑制IL-18表达在逆转MS心肌纤维化发生发展中起重要作用。
Objective To investigate the mechanism of felodipine in reversing metabolic syndrome (MS) myocardial fibrosis. Methods Thirty-three male Wistar rats were randomly divided into control group (n = 12) and metabolic syndrome group (n = 21). MS animal models were established by using 10% fructose for 8 months. Nine rats with metabolic syndrome Give felodipine. The content of collagen in left ventricular myocardium was detected by picric acid Sirius Red staining, and the expression of interleukin-18 (IL-18) mRNA was detected by RT-PCR. Results Compared with the control group, the content of collagen in left ventricular myocardium of MS rats increased (P <0.01) and the level of IL-18 mRNA increased (P <0.01). All the above abnormalities were relieved after treatment with felodipine. Conclusion Felodipine may play an important role in reversing the occurrence and development of myocardial fibrosis by inhibiting the expression of IL-18.
