鬼臼酰肼哌啶腙氮氧自由基(GP_1)和鬼臼乙叉甙(VP_(16))对L_(7712)细胞的增殖有明显的抑制作用,且有时间依赖性。在0.04~20mg/L范围,GP_1增加有丝分裂指数(MI)的作用与VP_(16)降低MI的作用均和剂量相关。5mg/LGP_1处理细胞12h时MI达峰值,为对照的8倍;而VP_(16)组的MI仅为对照的1/3。VP_(16)可对抗GP_1增高MI的作用。用GP_1或VP_(16)24h时细胞死亡、崩解增多。两药对S_(180),AH,L_(1210)和L_(7712)细胞DNA合成均有不同程度的抑制作用。
Peptidyl hydrazide, piperidine nitroxide (GP_1) and etoposide (VP_(16)) significantly inhibited the proliferation of L_(7712) cells in a time-dependent manner. In the range of 0.04 to 20 mg/L, the effect of increasing the mitogenic index (MI) of GP_1 was both dose-dependent and the effect of VP_(16) decreased MI. The MI reached a peak at 8 h after treatment with 5 mg/LGP_1 for 8 h, whereas it was only 1/3 of that for the control in the VP_16 group. VP_(16) can increase the effect of MI on GP_1. The cell death and disintegration increased with the use of GP 1 or VP 16 at 24 hours. Both drugs had different degrees of inhibition on DNA synthesis of S_ (180), AH, L_ (1210) and L_ (7712) cells.
