目的:探讨二鳖散对大鼠Walker-256肝癌肿瘤生长及血清血管内皮生长因子(VEGF)、内皮抑素(Endostatin)表达的影响,从抗血管生成角度探讨二鳖散的抗癌作用。方法:采用Walker-256癌肉瘤腹水接种Wistar大鼠制作肝癌模型,随机分为二鳖散低、中、高剂量(1.25,2.5,5g·kg-1·d-1)组、5-氟脲嘧啶75mg·kg-1组及模型组。于造模的第9天开始灌胃,每日1次,共21d。检测各组大鼠瘤重、肿瘤抑制率,ELISA法检测各组大鼠血清VEGF,endostatin的表达,计算比较各组VEGF/endostatin的比值。结果:二鳖散中、高剂量组和5-氟脲嘧啶组大鼠瘤重减轻,其血清VEGF表达量降低,Endostatin表达量升高,VEGF/endostatin比值降低,与模型组比较,差异有显著性(P<0.05或P<0.01)。结论:二鳖散能抑制大鼠Walker-256肝癌的生长,其机制可能与调控VEGF/endostatin的失衡而抑制肿瘤血管生成有关。
Objective: To investigate the effect of Erhuang Powder on tumor growth, serum vascular endothelial growth factor (VEGF) and endostatin expression of rat Walker-256 hepatoma, and to discuss the anti-cancer effect of Eryu powder from the perspective of anti-angiogenesis. METHODS: Wistar rats were inoculated with Walker-256 carcinoma sarcoma ascites to make a liver cancer model. They were randomly divided into two groups: low, medium and high dose (1.25, 2.5, 5 g·kg-1·d-1) groups, 5-fluorourea. Pyrimidine 75mg·kg-1 group and model group. On the 9th day of modeling, gavage was started once daily for 21 days. The tumor weight and tumor inhibition rate of each group were detected. The expression of VEGF and endostatin in serum of each group was detected by ELISA. The ratio of VEGF/endostatin in each group was calculated and compared. RESULTS: The tumor weight of rats in the middle and high dose group and 5-fluorouracil group was decreased. The serum VEGF expression was decreased, Endostatin expression was increased, and the ratio of VEGF/endostatin was decreased. Compared with the model group, the difference was significant. Sex (P<0.05 or P<0.01). Conclusion: Dijusan can inhibit the growth of Walker-256 hepatoma in rats, and its mechanism may be related to the regulation of VEGF/endostatin imbalance and inhibition of tumor angiogenesis.
