背景:近来的研究发现骨髓间充质干细胞(mesenchymalstemcells,MSCs)对缺血性脑血管病有治疗作用,接受细胞移植的脑梗死区超微结构的观察可进一步证实该作用。目的:观察接受MSCs局部注射移植的大鼠脑梗死区超微结构的变化,以了解MSCs移植对梗死区超微结构的影响。设计:随机对照实验研究。单位:解放军第三军医大学西南医院神经病学中心。材料:实验于2002-04/2002-09在解放军第三军医大学动物所及中心实验室完成。实验动物为健康幼年及成年Wistar大鼠12只,雌雄不限。随机分为移植组6只和对照组6只。干预:建立大鼠大脑中动脉栓塞模型,将体外分离培养的大鼠MSCs注射至大鼠脑梗死区,两组大鼠于移植后第4周电镜观察大鼠脑梗死区超微结构变化。主要观察指标:移植组及对照组大鼠脑梗死区超微结构变化。结果:接受移植的梗死区可见外形幼稚的细胞存活,并围绕在神经元周围,梗死区神经元胞浆中出现了丰富的游离核糖体,而对照组大鼠梗死区神经元胞核固缩,并有嗜神经细胞现象。结论:大鼠MSCs移植对缺血性脑损伤具有一定修复作用。
BACKGROUND: Recent studies have found that mesenchymal stem cells (MSCs) have a therapeutic effect on ischemic cerebrovascular disease. The observation of the ultrastructure of cerebral infarction area after cell transplantation can further confirm this effect. OBJECTIVE: To observe the ultrastructural changes of cerebral infarction in MSCs receiving local injection in order to understand the effect of MSCs transplantation on the ultrastructure of infarct area. Design: Randomized controlled trial. Unit: Southwest Hospital, Third Military Medical University PLA Center of Neurology. MATERIALS: The experiment was performed at the Institute of Zoology and Central Laboratory, Third Military Medical University of Chinese PLA from April 2002 to September 2002. Experimental animals for healthy young and adult Wistar rats 12, male or female. Randomly divided into transplantation group 6 and control group 6. Intervention: The rat model of middle cerebral artery occlusion was established. The rat MSCs isolated from in vitro were injected into the cerebral infarction area of rats. The ultrastructural changes of cerebral infarction in rats were observed under the electron microscope at the 4th week after transplantation. MAIN OUTCOME MEASURES: Ultrastructural changes in cerebral infarction area in transplantation and control groups. Results: Infarct cells in the infarcted area showed the survival of immature cells. Around the neurons, abundant free ribosomes appeared in the cytoplasm of neurons in the infarct area. In the control group, the nuclei of the neurons in the infarct area were pyknotic, And there are phenomena of addicted nerve cells. Conclusion: Transplantation of rat MSCs can repair ischemic brain injury.
