目的检测在白细胞介素-6(IL-6)环境培养下前列腺癌细胞微核糖核酸-21(miR-21)的表达,细胞的增殖能力.分析 IL-6和 miR-21对前列腺癌细胞的作用及意义.方法 RT-PCR 检测 miR-21的表达情况.CCK-8检测细胞增殖能力.通过 western blot 检测 PDCD4和 Bcl-2的表达情况.结果 IL-6诱导 miR-21的高表达,miR-21靶向 PDCD4、Bcl-2等下游基因,从而促进 PC-3细胞增殖.结论 IL-6通过调控 miR-21高表达有可能在前列腺癌中起重要作用.“,”Objective To detect the different expression of microRNA-21 aming prostate carcinoma cells in exposure to distinct concentration of interleukin-6 andits effcet on cell proliferation. Methods RT-PCR was used to detect the expression of microRNA-21 among prostate carcinoma cells exposed to distinct concentration of interleukin-6. CCK-8 was applied to check the capacity of cell proliferation. Expressions of Bcl-2 and PDCD4 were detected by western blot. Results IL - 6 enhanced high expression of miR- 21 and miR-21 regulated the target genes of PDCD4 and Bcl-2, further increased the proliferation of PC3 cells . Conclusion IL-6 promoted miR-21 expression,which might play a critical role in prostate cancer.
