目的 研究 艹猪 苓多糖抗小鼠HepA瘤细胞的可能机制。方法 采用免疫组化方法研究 艹猪苓多糖对小鼠HepA瘤细胞P16、Rb基因及TNF -α表达影响。结果 艹猪 苓多糖作用后 ,小鼠HepA瘤细胞P16基因表达蛋白 (P16蛋白 )表达显著增强 (P <0 .0 1) ;小鼠HepA瘤细胞Rb基因表达蛋白(P10 5 -Rb蛋白 )表达增强 (P <0 .0 5 ) ,小鼠HepA瘤细胞TNF -α表达显著增强 (P <0 .0 1)。结论 艹猪 苓多糖对小鼠HepA瘤细胞P16、Rb基因及TNF -α表达有激活作用。对抑癌基因P16、Rb基因及TNF -α表达的激活可能是艹猪 苓多糖抗肿瘤的普遍机制之一。
Objective To study the possible mechanism of Polyporus umbellatus polysaccharide against HepA tumor cells in mice. Methods Immunohistochemical method was used to study the effect of Polysaccharide from Polyporus umbellatus on the expression of P16, Rb gene and TNF-α in HepA tumor cells of mice. Results After Polyporus umbellatus Polysaccharide treatment, the expression of P16 protein in HepA tumor cells was significantly increased (P <0.01). The expression of Rb gene protein (P10 5 -Rb protein) in HepA tumor cells was significantly increased (P <0.05). The expression of TNF-α in HepA tumor cells was significantly increased (P <0.01). Conclusion Polyporus umbellatus polysaccharide on mouse HepA tumor cells P16, Rb gene and TNF-α expression activation. The activation of tumor suppressor gene P16, Rb gene and TNF-α expression may be one of the universal mechanisms of anti-tumor of Polyporus umbellatus polysaccharide.
