目的探讨胃癌组织中调节性T细胞浸润和缺氧诱导因子-1α(HIF-1α)的表达及联合检测对预后的预测价值。方法收集2006年12月至2007年12月90例胃癌患者临床及随访资料。免疫组织化学染色检测90例胃癌及癌旁组织内Foxp3和HIF-1α的表达。结果胃癌组织内Foxp3计数显著高于癌旁组织(10.93±7.49vs.4.10±2.03,t=9.19,P<0.01);胃癌组织内HIF-1α评分显著高于癌旁组织(6.48±3.09vs.2.87±1.72,t=10.80,P<0.01)。胃癌组织内Foxp3和HIF-1α的表达呈显著正相关(r=0.661,P<0.001);HIF-1α表达与肿瘤浸润深度(P=0.015)和肿瘤临床分期(P=0.001)相关,Foxp3表达与肿瘤临床分期(P=0.012)相关;Foxp3和HIF-1α均低表达的27例患者中位生存期显著长于Foxp3和HIF-1α均高表达的41例患者(P=0.005)。结论联合检测Foxp3和HIF-1α的表达可有效的预测胃癌预后。
Objective To investigate the expression of regulatory T cell infiltration and hypoxia-inducible factor-1α (HIF-1α) in gastric cancer and their prognostic value. Methods The clinical and follow-up data of 90 gastric cancer patients from December 2006 to December 2007 were collected. Immunohistochemical staining was used to detect the expression of Foxp3 and HIF-1α in 90 cases of gastric cancer and paracancerous tissues. Results The Foxp3 counts in gastric cancer tissues were significantly higher than those in adjacent tissues (10.93 ± 7.49 vs. 4.10 ± 2.03, t = 9.19, P <0.01). The scores of HIF-1α in gastric cancer tissues were significantly higher than those in adjacent tissues (6.48 ± 3.09 vs. 2.87 ± 1.72, t = 10.80, P <0.01). There was a significant positive correlation between the expression of Foxp3 and HIF-1α in gastric cancer (r = 0.661, P <0.001). The expression of HIF-1α was correlated with the depth of tumor invasion (P = 0.015) (P = 0.012). The median survival time of 27 patients with low expression of Foxp3 and HIF-1α was significantly longer than that of 41 patients with high expression of Foxp3 and HIF-1α (P = 0.005). Conclusion The combined detection of Foxp3 and HIF-1α expression can effectively predict the prognosis of gastric cancer.
