目的探讨人类白细胞抗原DR(human leucocyte antigen DR,HLA-DR)和Ⅱ类反式激活因子(classⅡtransactivator,CⅡTA)在肝细胞癌(hepatocellular carcinoma,HCC)患者癌组织与癌周肝组织中的表达水平及其意义。方法用逆转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测33例乙型肝炎相关性肝细胞癌患者手术切除的肝癌组织和癌周肝组织中CⅡTA mRNA的表达状况,并用免疫组织化学染色检测肝癌组织和癌周肝组织中HLA-DR和CⅡTA蛋白的表达水平。结果癌周肝组织中HLA-DR蛋白、CⅡTA蛋白与mRNA表达阳性检出率均为100%,肝癌组织中HLA-DR蛋白与CⅡTA蛋白表达阳性检出率分别为24.2%和33.3%,CⅡTA mRNA未检测到阳性表达,两者间差异显著(P<0.05);33例患者的肝癌细胞中均缺乏HLA-DR与CⅡTA蛋白表达,而癌周组织肝细胞中均有不同程度HLA-DR与CⅡTA蛋白表达;组织中HLA-DR表达水平与CⅡTA表达水平呈明显正相关(rs=0.9085,P<0.05)。结论肝癌细胞缺乏HLA-DR表达,与其不表达CⅡTA有关,并可能在肝癌细胞免疫逃逸中起重要作用。
Objective To investigate the expression of human leukocyte antigen DR (HLA-DR) and class Ⅱ transactivator (CⅡTA) in the cancer tissues and peritumor liver tissues of patients with hepatocellular carcinoma (HCC) And its significance. Methods The expression of CⅡTA mRNA in 33 cases of hepatocellular carcinoma and peritumor liver tissues of 33 cases of hepatocellular carcinoma with hepatocellular carcinoma was detected by reverse transcription-polymerase chain reaction (RT-PCR) Immunohistochemical staining was used to detect the expression of HLA-DR and CⅡTA in hepatocellular carcinoma and peritumorous liver. Results The positive rates of HLA-DR protein and CⅡTA protein and mRNA expression in peri-hepatic tissue were all 100%. The positive rates of HLA-DR and CⅡTA in hepatocellular carcinoma tissues were 24.2% and 33.3%, respectively. CⅡTA mRNA No significant difference was found between the two groups (P <0.05). The expression of HLA-DR and CⅡTA protein in all the 33 cases of hepatocellular carcinoma cells was absent. However, the expression of HLA-DR and CⅡTA in hepatocellular carcinoma tissues was different The expression of HLA-DR in tissues was positively correlated with the expression of CⅡTA (rs = 0.9085, P <0.05). Conclusion The lack of HLA-DR expression in HCC cells may be related to the lack of expression of CⅡTA and may play an important role in the immune escape of HCC cells.
