目的研究17β-雌二醇(E2)对成年雌兔关节软骨细胞的作用,探查一氧化氮(NO)是否在其中发挥中介作用。方法不同浓度E2(10-12M~10-3M)与软骨细胞共同孵育后一定时间后检测结果。用化学比色法分别检测软骨细胞增殖与能量代谢、细胞间质中胶原含量以及培养液中一氧化氮合酶(NOS)活力;硝酸还原酶法检测培养液中NO含量。结果在10-12M~10-6M剂量组,E2对软骨细胞增殖与能量代谢起剂量依赖性的抑制作用(P<0.01);在10-12M~10-9M剂量组,E2对软骨细胞合成胶原无显著性作用(P>0.05);在10-8M~10-6M剂量组,E2起剂量依赖性的抑制作用(P<0.05);在10-7M和10-6M剂量组,E2对细胞培养液中NOS活力及NO含量均无显著性作用(P>0.05);在10-5~10-3M剂量组,E2能显著性地增加培养液中NOS活力及NO的含量(P<0.05)。结论E2对软骨细胞具有“直接”调节作用,该作用与雌激素浓度呈显著性相关。NO可能是雌激素对软骨发挥抑制作用的中介因素之一。
Objective To investigate the effect of 17β-estradiol (E2) on articular chondrocytes in adult female rabbits and to explore whether nitric oxide (NO) plays an intermediary role. Methods Different concentrations of E2 (10-12M ~ 10-3M) were incubated with chondrocytes after a certain period of time after the test results. The proliferation and energy metabolism of chondrocytes, the content of collagen in interstitial cells and the activity of nitric oxide synthase (NOS) in culture medium were detected by chemical colorimetric method respectively. The content of NO in culture medium was detected by nitrate reductase method. Results In the dose of 10-12M ~ 10-6M, E2 inhibited the proliferation and energy metabolism of chondrocytes in a dose-dependent manner (P <0.01). In the dose of 10-12M ~ 10-9M, E2 inhibited the collagen synthesis of chondrocytes (P <0.05). In the dose of 10-8M ~ 10-6M, E2 dose-dependently inhibited the proliferation of cells (P <0.05); in 10-7M and 10-6M dose groups, (P> 0.05). In the 10-5 ~ 10-3M dose group, E2 significantly increased NOS activity and NO content in the medium (P <0.05). Conclusion E2 has a “direct” regulatory effect on chondrocytes, which is significantly correlated with estrogen concentration. NO may be one of the mediators of the inhibitory effect of estrogen on cartilage.
