目的:建立一种准确度和灵敏度高,重现性好的山楂叶提取物中牡荆素鼠李糖苷的高效液相色谱测定法。方法:色谱柱为Zorbax SB-C18(250mm×4.6 mm,5μm),流动相A为乙腈,B为水溶液,梯度洗脱。0~5m in,流动相A由5%~10%,B由95%~90%;5~10m in,A由10%~18%,B由90%~82%;10~30m in,A保持18%,B保持82%,流速为1.0mL/m in,检测波长334 nm,柱温30℃。结果:牡荆素鼠李糖苷在4.296~64.440μg/mL浓度范围内与峰面积值线性关系良好(r=1),平均回收率为99.19%,RSD=1.94%。结论:建立的含量测定方法简便、准确、灵敏,可用于山楂叶提取物中牡荆素鼠李糖苷的测定。
Objective: To establish a HPLC method for the determination of vitexin rhamnoside in Hawthorn leaf extract with high accuracy and sensitivity. METHODS: The chromatographic column was Zorbax SB-C18 (250 mm×4.6 mm, 5 μm). The mobile phase A was acetonitrile and B was an aqueous solution with gradient elution. 0~5m in, mobile phase A from 5% to 10%, B from 95% to 90%, 5 to 10m in, A from 10% to 18%, B from 90% to 82%, 10 to 30m in, A Maintained 18%, B remained 82%, flow rate 1.0 mL/min, detection wavelength 334 nm, column temperature 30°C. RESULTS: The linear relationship between vitexin rhamnoside and peak area values was good in the concentration range of 4.296~64.440μg/mL (r=1). The average recovery rate was 99.19% and the RSD was 1.94%. Conclusion: The established method is simple, accurate and sensitive. It can be used for the determination of vitexin rhamnoside in hawthorn leaf extract.
