目的:研究中药胃肠舒对糖尿病(diabetes mellitus,DM)大鼠胃窦干细胞生长因子及其受体(stemcellfactor-kit,SCF-kit)和神经递质的影响,探讨胃肠舒促胃肠运动的机制。方法:标准SD大鼠随机分为:正常对照组、DM组、DM+胃肠舒Ⅰ组、DM+胃肠舒Ⅱ组。流式细胞仪检测各组大鼠胃窦组织中Cajal间质细胞(interstitial cells of Cajal,ICC)的数量,RT-PCR、Western blot分别测定SCF-kit mRNA及蛋白水平上的表达,免疫组化法、激光共聚焦显微镜分别检测神经递质及Ca2+的变化。结果:胃肠舒可增加DM大鼠胃窦ICC的数量,上调SCF-kit mRNA及蛋白表达,降低NO的表达,Ach、Ca2+表达增高。结论:胃肠舒可能通过改变SCF-kit和神经递质的表达,改善了ICC,从而促进胃肠运动。
Objective: To study the effect of weizheshu on gastric anastomosis and its receptor (SCF-kit) and neurotransmitter in diabetic mellitus (DM) rats and to explore the effect of gastrointestinal comfort and promoting gastrointestinal motility Mechanisms. Methods: SD rats were randomly divided into normal control group, DM group, DM + GI group, DM + GI group. The number of Cajal interstitial cells of Cajal (ICC) in gastric antrum was detected by flow cytometry. The expression of SCF-kit mRNA and protein was detected by RT-PCR and Western blot, respectively. Immunohistochemistry Method and laser confocal microscopy were used to detect neurotransmitter and Ca2 + changes. Results: Gastrointestinal Shu can increase the number of ICC in gastric antrum, increase the expression of SCF-kit mRNA and protein, decrease the expression of NO and increase the expression of Ach and Ca2 + in DM rats. Conclusion: Gastrointestinal Shu can improve the ICC by changing the expression of SCF-kit and neurotransmitter, so as to promote gastrointestinal motility.
