TRAIL(TNF relatedapoptosis inducingligand ,简称TRAIL或Apo 2L)是肿瘤坏死因子家族的新成员 ,体外研究表明其具有特异性杀伤肿瘤细胞的功能 .为了进一步探索TRAIL的生物学功能及其临床应用前景 ,首次从中国正常人外周血淋巴细胞中扩增了TRAIL胞外功能区(sTRAIL) ,并将其插入原核表达载体 ,转化大肠杆菌后 ,可溶型重组蛋白 (rsTRAIL)获得了高效稳定表达 ,其产量达到菌体总蛋白的 30 %左右 .体外研究表明 ,亲和层析纯化后的重组蛋白在 0 .1~ 1 μg/mL的浓度范围内 ,能显著诱导所试全部 7种肿瘤细胞株细胞凋亡 ,但不能诱导正常鼠成纤维细胞株细胞凋亡 ;进一步研究表明 ,rsTRAIL对贲门癌、乳腺癌和恶性胸腺瘤等原代细胞也具有强烈的杀伤作用 ,但不能杀伤正常人外周血淋巴细胞 .此实验结果为rsTRAIL特异性地诱导肿瘤细胞凋亡的生物学作用提供了新的实验证据 ,具有重要的临床应用价值 .
