目的探讨沙门菌属环介导等温扩增(loop-mediated isothermal amplification,LAMP)方法在食品卫生检验中的应用价值。方法利用文献报道的LAMP检测方法和分离培养法分别对135份食品标本进行沙门菌检测,评价LAMP法和分离培养法的一致性。结果 LAMP方法检出14份食品标本沙门菌阳性,阳性率为10.4%,电泳和加荧光染料染色后均能判断结果;分离培养法检出6份食品标本沙门菌阳性,阳性率为4.4%。LAMP法检出阳性率高于培养法(P=0.021)。LAMP方法可在24 h内从食品中检测出沙门菌,而分离培养法检出沙门菌需要4~7 d,LAMP检测方法更快速。结论沙门菌属LAMP方法检测阳性率高于分离培养法,LAMP方法可用于食品中沙门菌的核酸检测。
Objective To investigate the value of loop-mediated isothermal amplification (LAMP) method in food hygiene inspection of Salmonella. Methods LAMP detection method and isolation culture method were used to detect Salmonella in 135 food samples, and to evaluate the consistency of LAMP method and isolation culture method. Results The positive rate of Salmonella in 14 food samples detected by LAMP method was 10.4%. The results of electrophoresis and fluorochrome staining showed that Salmonella were positive in 6 food samples and the positive rate was 4.4%. The positive rate of LAMP was higher than that of culture (P = 0.021). The LAMP method can detect Salmonella from food within 24 h, while Salmonella detected by isolation culture method takes 4 ~ 7 days, and LAMP detection method is faster. Conclusion The detection rate of Salmonella genus LAMP is higher than that of isolation culture method. The LAMP method can be used for nucleic acid detection of Salmonella in food.
