利用RT-PCR技术,从被白粉菌诱导的小麦-簇毛麦6VS/6AL易位系中获得病程相关蛋 白1(TaPr-1)。DNA克隆.该基因具有编码164氨基酸的开放阅读框,其中包含24个氨基酸构 成的信号肽和140个氨基酸组成的成熟肽(MW为15.1kD).经蛋白结构比较分析,发现其与 植物中已分离的多种 PK类蛋白具一定的同源性。 Northern杂交显示,易位系被诱导 12小时 左右,该基因转录物累积最多,其在抗病易位系和感病亲本扬5中表达水平有明显差异。 Southern杂交表明在小麦基因组中该基因的拷贝数多于1个,且该基因在扬5和易位系中表 现多态性,表明6VS上有可能携带该基因。
Pathogen-related protein 1 (TaPr-1) was obtained from the 6VS / 6AL translocation line induced by powdery mildew in wheat using RT-PCR. DNA clone. This gene has an open reading frame (ORF) coding 164 amino acids and contains a signal peptide consisting of 24 amino acids and a mature peptide consisting of 140 amino acids (MW 15.1 kD). Compared with the protein structure, we found that it has some homology with many kinds of PK proteins that have been isolated in plants. Northern blotting showed that the translocation line was induced for about 12 hours and the transcripts of this gene accumulated the most, which showed a significant difference in expression level between disease-resistant translocation line and susceptible parent Yang5. Southern blotting showed that there were more than one copy number of this gene in wheat genome, and the gene showed polymorphism in Yang 5 and the translocation line, indicating that it is possible to carry this gene in 6VS.
