目的探讨青白散抗炎作用及对肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)在大鼠慢性损伤骨骼肌中表达的影响。方法采用小鼠二甲苯性耳肿胀、大鼠蛋清性足肿胀和棉球性肉芽肿等多种炎症模型,观察青白散的抗炎作用;建立慢性软组织损伤动物模型,检测大鼠慢性软组织损伤后修复过程中TNF-α和IL-6水平的动态变化,并分析其相关性。结果 (1)青白散可明显抑制小鼠二甲苯性耳肿胀,显著抑制大鼠蛋清性足肿胀和棉球性肉芽组织增生。(2)模型组骨骼肌中TNF-α和IL-6的含量均较正常对照组显著增高(P<0.01)。(3)青白散组在不同时间点的骨骼肌中TNF-α和IL-6的含量均较模型对照组显著降低(P<0.01),与云南白药组比较差异无统计学意义(P>0.05)。结论青白散有明显的抗炎作用,能显著抑制大鼠慢性损伤骨骼肌中致炎因子TNF-α和IL-6的含量增高,该作用可能是青白散治疗慢性软组织损伤的主要机理之一。
Objective To investigate the anti-inflammatory effect of Qingbai San and its effect on the expression of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in chronic injured skeletal muscle of rats. Methods Various inflammatory models such as xylene-induced ear swelling in mice, egg-like foot swelling in rats and granuloma of cotton ball were used to observe the anti-inflammatory effects of Qingbai San. A chronic soft tissue injury animal model was established and the chronic soft tissue injury The dynamic changes of TNF-α and IL-6 during the repair process were analyzed and their correlations were analyzed. Results (1) Qingbai San can significantly inhibit xylene ear swelling in mice, significantly inhibited rat egg white paw swelling and granulosa granulation tissue hyperplasia. (2) The levels of TNF-α and IL-6 in model group were significantly higher than those in normal control group (P <0.01). (3) The contents of TNF-α and IL-6 in skeletal muscle of Qingbai powder group were significantly lower than those of model control group at different time points (P <0.01), but there was no significant difference between them ). Conclusion Qingbai San has obvious anti-inflammatory effect, which can significantly inhibit the increase of inflammatory cytokines TNF-α and IL-6 in rats with chronic injury. This effect may be one of the main mechanisms of Qingbai San in treating chronic soft tissue injury.
