目的探讨C57BL/6J小鼠继发腭发育过程中,甲状腺转录因子-2(TTF-2)和重组人转化生长因子β3(TGFβ3)的表达及其相互作用规律。方法于妊娠12、13、14d分别断颈处死各6只孕鼠,无菌条件取出胚胎,解离腭突。应用免疫组织化学染色和Western印记方法检测TTF-2和TGFβ3在腭突中嵴上皮细胞的表达。结果在G57BL/6J小鼠胚胎的腭突形成、上抬和融合的发育过程中均有TTF-2的表达,以13d为最高,免疫组织化学和Western印记结果一致。TGFβ3在腭突形成时,腭突中嵴上皮细胞未检测到其表达。腭突上抬时,首次在腭中山嵴上皮细胞中发现TGFβ3表达,两侧腭突融合时,TGFβ3在腭中嵴上皮中的表达最明显,当腭突融合完成后,TGFβ3在腭中嵴上皮中表达减少。结论 TTF-2和TGFβ3两种细胞因子均在腭突中嵴上皮细胞中表达,表明与其分化转归相关。
Objective To investigate the expression of TTF-2 and TGFβ3 during secondary palatal development in C57BL / 6J mice and its interaction. Methods Six pregnant rats were sacrificed on the 12th, 13th, and 14th day of gestation. The embryos were removed aseptically and the palate was dissociated. Immunohistochemical staining and Western blotting were used to detect the expression of TTF-2 and TGFβ3 in the midgut epithelium. Results The expression of TTF-2 in the process of development, uplift and fusion of the palatal epithelium of G57BL / 6J mouse embryos was highest, which was the highest at 13d. The results of immunohistochemistry and Western blot were consistent. TGFβ3 in the formation of palatal, the palatal crest epithelial cells did not detect its expression. The expression of TGFβ3 was found in the palatal crest epithelial cells for the first time when the palatal protrusion was raised. The expression of TGFβ3 in the palatal midgut epithelium was the most obvious when both palatal fossa were fused. When the palatal fossa was fused, Reduced expression. Conclusion Both TTF-2 and TGFβ3 cytokines are expressed in the midgut epithelial cells of the palatal process, indicating that they are related to their differentiation and metastasis.
