目的:探讨苗药生仙汤对大鼠马尾损伤(cauda equina injury,CEI)后脑源性神经营养因子(brain derived neurophic factor,BDNF)及其mRNA(BNDF mRNA)表达的影响。方法:成年雌性SD大鼠60只,随机分成正常组、对照组、苗药组和西药组。采用钳夹损伤法复制大鼠CEI模型,正常组大鼠不做任何处理,实验组大鼠术后4h即予生理盐水、苗药生仙汤煎剂和甲钴胺溶液灌胃。各组动物分别于术后1d、7d和14d处死,处死前采用BBB评分法评估大鼠神经功能情况,取材后应用实时荧光定量PCR法和免疫组织化学染色结合图像定量分析法分别检测大鼠骶髓中BDNF蛋白及其mRNA的表达水平。结果:术后1d,对照组、苗药组和西药组BDNF蛋白表达明显高于正常组(P<0.05),西药组BDNF蛋白及其mRNA表达明显高于对照组和苗药组(P<0.05),而对照组和苗药组BDNF蛋白及其mRNA表达差异无统计学意义(P>0.05);术后7d,实验组BDNF蛋白及其mRNA表达均有所上升,苗药组和西药组表达明显高于对照组(P<0.05),而苗药组和西药组表达差异无统计学意义(P>0.05);术后14d,实验组BDNF蛋白及其mRNA表达均呈下降趋势,但仍高于术后1d时水平(P<0.05),而各组之间表达比较情况同术后7d。BBB评分显示实验组大鼠术后神经功能均趋于恢复,但苗药组和西药组大鼠恢复速度明显快于对照组(P<0.05),苗药组和西药组大鼠差异无统计学意义(P>0.05)。结论:苗药生仙汤可以增加CEI大鼠骶髓BDNF及其mRNA的表达,从而促进神经修复与再生,抑制神经元凋亡。
Objective: To investigate the effect of Miaosheng Shengxian decoction on the expression of brain derived neurophic factor (BDNF) and its mRNA (BNDF mRNA) after Cauda equina injury (CEI) in rats. Methods: 60 adult female Sprague-Dawley rats were randomly divided into normal group, control group, Miao drug group and western medicine group. The rat model of CEI was duplicated by the clamp injury method. Rats in the normal group were given no treatment. Rats in the experimental group were given normal saline 4 hours after the operation, and Miao Yin Sheng Xian Tang decoction and methylcobalamin solution were given orally. Animals in each group were sacrificed on the 1st, 7th and 14th day after operation. The BBB score was used to assess the neurological function of the rats before sacrifice. Real-time quantitative PCR and immunohistochemistry were used to detect the neurological function of rats. The expression of BDNF protein and its mRNA in the marrow. Results: At 1 day after operation, the BDNF protein expression in the control group, Miao drug group and western medicine group was significantly higher than that in the normal group (P <0.05). The protein and mRNA expression of BDNF in the western medicine group were significantly higher than those in the control group and the Miao drug group ), While there was no significant difference in BDNF protein and mRNA expression between control group and Miao drug group (P> 0.05). On the 7th day after operation, the expression of BDNF protein and its mRNA in experimental group increased (P <0.05), while there was no significant difference between Miao drug group and western medicine group (P> 0.05). On the 14th day after operation, the expression of BDNF protein and mRNA in the experimental group showed a decreasing trend, but still high (P <0.05) at 1 day after operation, while the expression of each group was compared with that of 7 days after operation. BBB score showed that the neurological function of rats in experimental group tended to recover, but the recovery speed of Miao drug group and western medicine group was significantly faster than that of control group (P <0.05), there was no significant difference between Miao drug group and western medicine group Significance (P> 0.05). Conclusion: Miaosheng Shengxian Decoction can increase the expression of BDNF and its mRNA in sacral sac of CEI rats, and promote the repair and regeneration of neurons and inhibit the apoptosis of neurons.
