目的:探讨脑蛋白水解物注射液中肽含量的测定方法并试图比较不同厂家产品的质量差异。方法:采用盐酸水解多肽方法,利用AccQ-Tag氨基酸衍生技术,使用Kromasil100-5C18色谱柱(250mm×4.6mm,5μm)进行液相色谱分析,水解后游离氨基酸总量扣除水解前氨基酸总量即为肽含量。结果:各氨基酸测定结果具有良好线性(r=0.9997~0.9999,n=6),肽平均回收率为101.0%(n=9)。结论:本方法准确可行,可作为药品中肽含量测定的方法补充和完善。
Objective: To explore the method of determination of peptide in brain protein hydrolyzate injection and try to compare the quality differences of different manufacturers’ products. Methods: Hydrochloric acid hydrolyzed peptides were analyzed by AccQ-Tag amino acid derivatization with Kromasil100-5C18 column (250 mm × 4.6 mm, 5 μm). The total amount of free amino acids after hydrolysis was calculated by subtracting the total amount of amino acids before hydrolysis Peptide content. Results: The results of each amino acid showed good linearity (r = 0.9997 ~ 0.9999, n = 6). The average recovery rate of peptide was 101.0% (n = 9). Conclusion: This method is accurate and feasible, and can be used as a method to determine the content of peptide in medicine to supplement and improve.
