目的建立HPLC-DAD同时测定中药知母中芒果苷、知母皂苷BⅡ的方法。方法采用HPLC法,色谱柱为Hypersil GOLD色谱柱(250 mm×4.6 mm,5μm),乙腈-0.01%磷酸水梯度洗脱,流速1 mL/min,检测波长191 nm和258 nm,柱温:30℃。结果芒果苷的线性范围在2.944~6.624μg,r=0.999 8;平均回收率为101.06%,RSD=1.61%(n=6)。知母皂苷BⅡ的线性范围在18.080~40.680μg,r=0.999 4;平均回收率为103.43%,RSD=1.43%(n=6)。结论 3批样品测定结果表明,该方法简便准确,可用于知母中芒果苷、知母皂苷BⅡ2种成分的含量测定。
Objective To establish a method for the simultaneous determination of mangiferin and timosaponin B Ⅱ in Zhimu, a traditional Chinese medicine by HPLC-DAD. Methods HPLC was performed on a Hypersil GOLD column (250 mm × 4.6 mm, 5 μm) with a gradient of acetonitrile to 0.01% phosphoric acid. The flow rate was 1 mL / min. The detection wavelength was 191 nm and 258 nm. The column temperature was 30 ℃. Results The linear range of mangiferin was 2.944 ~ 6.624μg, r = 0.999 8; the average recovery was 101.06%, RSD = 1.61% (n = 6). The linear range of timosaponin BⅡwas 18.080 ~ 40.680μg, r = 0.999 4; the average recovery was 103.43%, RSD = 1.43% (n = 6). Conclusion The results of three batches of samples show that the method is simple and accurate and can be used to determine the content of two components of mangiferin and timosaponin B in Zhimu.
