目的:观察益气化瘀方对腰神经根压迫损伤后L4神经根组织Bax和Bcl-2的表达影响。方法:雄性SD大鼠(SPF级)40只按随机数字表法分为假手术组、模型组、弥可保组和益气化瘀方组,每组各10只。采用自制胶块压迫于大鼠右侧L4神经根背根节处,假手术组仅切开皮肤和椎旁肌。造模后假手术组和模型组给予生理盐水灌胃;弥可保组采用肌肉注射弥可保注射液;益气化瘀方组给予益气化瘀方煎剂灌胃,于造模10d后取右侧受压神经根。采用免疫组织化学法检测背根节神经细胞Bax和Bcl-2的蛋白表达情况;实时荧光定量聚合酶链反应法检测神经根组织Bax和Bcl-2的基因表达变化。结果:腰神经根压迫后,模型组的Bax蛋白和基因表达明显增高,其中基因表达与假手术组比较差异有统计学意义(P<0.01);而Bcl-2蛋白和基因表达明显减少,其中基因表达与假手术组比较差异有统计学意义(P<0.01)。益气化瘀方可减少Bax的蛋白和基因表达,其中基因表达低于模型组(P<0.01)。另外益气化瘀方可增加Bcl-2的蛋白表达,但对Bcl-2基因表达无明显影响。结论:腰神经根压迫模型可导致受压神经根组织Bax的过表达增加和Bcl-2的表达抑制。益气化瘀方可抑制神经根组织Bax的过表达;可增加Bcl-2的蛋白表达,但对其基因表达无明显影响。
Objective: To observe the effect of Yiqi Huayu Recipe on the expression of Bax and Bcl-2 in L4 nerve root tissue after lumbar nerve root compression injury. Methods: Forty male SD rats (SPF grade) were randomly divided into sham-operation group, model group, Mi-Ke-Bao group and Yiqi Huayu-Fang group, 10 in each group. Using self-made plastic block oppression in the rat right L4 nerve root dorsal root ganglia, sham operation group only cut the skin and paravertebral muscles. After modeling, sham operation group and model group were administrated with saline; intramuscular injection of Miqibao injection was administered to miconazole group; Yiqi Huayu decoction group was given intragastrically, Take the right compression nerve root. Immunohistochemistry was used to detect the protein expression of Bax and Bcl-2 in the dorsal root ganglion neurons. Real-time fluorescent quantitative polymerase chain reaction was used to detect the gene expression changes of Bax and Bcl-2 in the nerve root tissue. RESULTS: After compression of the lumbar nerve root, the expression of Bax protein and gene in model group was significantly higher than that in sham operation group (P <0.01), while the expression of Bcl-2 protein and gene was significantly decreased There was significant difference between gene expression and sham group (P <0.01). Yiqi Huayu Decoction can reduce Bax protein and gene expression, which gene expression was lower than the model group (P <0.01). In addition, Yiqi Huayu can increase the protein expression of Bcl-2, but had no significant effect on Bcl-2 gene expression. Conclusion: The lumbar nerve root compression model can lead to the overexpression of Bax and inhibition of Bcl-2 expression in the compressed nerve root tissue. Yiqi Huayu Recipe can inhibit Bax overexpression in nerve root tissue, increase the protein expression of Bcl-2, but have no obvious effect on its gene expression.
