目的:观察罗勒胶囊对大鼠移植性肝癌经肝动脉化疗栓塞术(TACE)后细胞中骨桥蛋白(OPN)和诱导性一氧化氮合酶(iNOS)表达的影响,探讨罗勒胶囊对肝肿瘤乏氧微环境的影响及意义。方法:建立大鼠移植性肝癌模型,10 d后将造模成功的大鼠随机分为5组。对TACE+生理盐水组和TACE+罗勒胶囊高、中、低剂量组4组大鼠进行TACE,并分别给予生理盐水10 mL.kg-1.d-1和罗勒胶囊300,150,75 mg.kg-1.d-1ig,对非TACE对照组大鼠只进行开腹操作,不进行TACE。给药10d后取出移植性肝癌病理标本,免疫组化法检测肿瘤组织中OPN和iNOS表达情况。结果:瘤重:行TACE的4组大鼠与非TACE组比较均明显减轻(P<0.05),TACE+罗勒胶囊高剂量组较TACE+生理盐水组显著减轻(P<0.05);OPN和iNOS阳性表达率:TACE+生理盐水组与非TACE组比均显著升高(P<0.01);TACE+罗勒胶囊高、中剂量组较TACE+生理盐水组显著降低(P<0.01);TACE+罗勒胶囊高剂量组较TACE+罗勒胶囊中、低剂量组阳性表达率低(P<0.01)。结论:中药罗勒胶囊能改善肝肿瘤局部的乏氧微环境并促进细胞凋亡,使TACE后肿瘤组织中OPN和iNOS表达降低,抑制大鼠移植性肝癌TACE术后肿瘤的复发。
Objective: To observe the effect of basil capsule on the expression of osteopontin (OPN) and inducible nitric oxide synthase (iNOS) in rat transplanted hepatocellular carcinoma after transcatheter arterial chemoembolization (TACE), and to investigate the effect of basil capsule on liver tumor The Influence and Significance of Hypoxic Microenvironment. Methods: The rat model of transplanted liver cancer was established. After 10 days, the successful model rats were randomly divided into 5 groups. Pathological specimens of transplanted hepatocellular carcinoma were removed 10 days after administration, and the expression of OPN and iNOS in tumor tissue was detected by immunohistochemistry. Results: Tumor weight: Compared with non-TACE group, the TACE group and the high-dose basil group significantly reduced the expression of OPN and iNOS (P <0.05) (P <0.01); TACE + basil high and medium dose groups were significantly lower than TACE + saline group (P <0.01); TACE + basil high dose group than TACE + In basil low dose group, the positive rate was lower (P <0.01). Conclusion: The traditional Chinese medicine Basil capsule can improve the local hypoxia microenvironment and promote the apoptosis of hepatocarcinoma, reduce the expression of OPN and iNOS in the tumor tissue after TACE, and inhibit the tumor recurrence after transcatheter arterial chemoembolization.
