目的:体外分离纯化人牙周膜干细胞,研究其体外向神经细胞分化的能力,为牙周组织工程提供可靠的种子细胞来源。方法:采用有限稀释法克隆分离纯化人牙周膜干细胞,将获得的第5代细胞用神经诱导体系连续培养,观察细胞形态变化;通过免疫组织化学染色技术检测诱导后神经胶质细胞标志物(GFAP)、神经元标志物(NSE)的表达。结果:人牙周膜细胞在体外诱导后出现了神经胶质样、神经元细胞分化的形态学改变,并有相应标志蛋白的表达,结论:人牙周膜干细胞在体外神经诱导体系作用下出现向神经细胞的分化。
OBJECTIVE: To isolate and purify human periodontal ligament stem cells in vitro and study their ability to differentiate into neurons in vitro, so as to provide a reliable source of seed cells for periodontal tissue engineering. Methods: The human periodontal ligament stem cells were isolated and purified by limited dilution method. The 5th passage cells were cultured continuously by neural induction system to observe the changes of cell morphology. The expression of glial cell markers GFAP), neuronal markers (NSE). RESULTS: After in vitro induction of human periodontal ligament cells, the morphological changes of glial and neuronal cell differentiation were observed and the expression of corresponding marker protein was observed. Conclusion: Human periodontal ligament stem cells appeared under the action of neural induction system in vitro Differentiation into nerve cells.
