目的建立固相萃取-高效液相色谱法测定青银注射液中蒿酮的分析方法。方法采用高效液相色谱法测定。色谱柱为Dikma Kromasil C18(250 mm×4.6 mm,5μm);流动相:乙腈-水(75∶25);检测波长:242 nm;柱温:30℃;体积流量:0.8 mL/min;进样量:10μL。供试品溶液的制备采用C18固相萃取小柱(甲醇10 mL活化,水10 mL平衡),用50%甲醇洗脱,弃去洗脱液,再用甲醇洗脱。结果蒿酮在0.220 5~3.527 2μg具有良好的线性关系(r=0.999 9),平均回收率为98.28%,RSD为0.35%。结论该方法简便、快速、准确,具有良好的重复性和回收率,可作为青银注射液中蒿酮的定量分析方法。
Objective To establish a solid phase extraction-high performance liquid chromatographic method for the determination of artenisia in Qingyin injection. Methods using high performance liquid chromatography. The column was Dikma Kromasil C18 (250 mm×4.6 mm, 5 μm); mobile phase: acetonitrile-water (75:25); detection wavelength: 242 nm; column temperature: 30°C; volume flow rate: 0.8 mL/min; Quantity: 10 μL. The test solution was prepared using a C18 solid-phase extraction cartridge (10 mL methanol activated, 10 mL water balanced), eluted with 50% methanol, discarded the eluent, and eluted with methanol. The results showed that there was a good linear relationship between 0.220 5 and 3.527 2 μg (r=0.999 9). The average recovery was 98.28% and the RSD was 0.35%. Conclusion The method is simple, rapid, accurate, and has good repeatability and recovery. It can be used as a quantitative analysis method of artemisinone in Qingyin injection.
