Objective: To investigate the role of endothelin in the pathogenesis of renal dysfunction in acute liver failure. Methods: The dynamic changes of liver and kidney function and endothelin level in rats with acute hepatic failure induced by thioacetamide (TAA) were observed. Results: With the aggravation of liver injury, renal function was also deteriorated (manifested by increased serum creatinine, decreased creatinine clearance, decreased urine output, decreased urinary sodium secretion, etc.), plasma endotoxin gradually increased, the most in 48h serious. With the above changes, endothelin activity in renal tissue gradually increased, reaching a peak at 48h. Plasma endothelin and creatinine clearance and creatinine clearance and renal tissue endothelin levels were significantly negative correlation, while plasma endothelin levels did not change significantly. CONCLUSION: Endothelin plays an important role in the development of renal dysfunction in acute liver failure, but it appears to have some relationship with endotoxin
