目的研究血管抑素体内抑制视网膜新生血管形成疗效及对血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法建立链脲佐菌素糖尿病大鼠模型,分A组(造模实验组),B组(造模对照组),每组36只,取正常SD大鼠36只作为C组(正常对照组)。予以玻璃体腔内注射血管抑素后观察视网膜HE染色结果及大鼠的视网膜VEGF的表达。结果 A、B组大鼠的血糖值明显高于同期C组;A组即注射血管抑素与B组即注射磷酸盐缓冲液(phosphate buffer solution,PBS)相比,视网膜的细胞排列有一定规则,组织水肿不明显;A组视网膜的VEGF水平明显低于B组,且都高于C组。结论血管抑素可明显降低糖尿病鼠视网膜组织的VEGF表达,能有效抑制糖尿病大鼠视网膜组织水肿及内部组织结构的改变。因此,血管抑素有潜在预防和治疗糖尿病性视网膜病变的价值。
Objective To investigate the effect of angiostatin on the inhibition of retinal neovascularization and its effect on the expression of vascular endothelial growth factor (VEGF) in vivo. Methods Streptozotocin-induced diabetic rats were divided into group A (model experimental group) and group B (model control group), 36 in each group. Thirty-six normal SD rats were used as group C (normal control group ). After intravitreal injection of angiostatin, retinal HE staining and retinal VEGF expression were observed. Results The blood glucose level of rats in groups A and B was significantly higher than that in group C at the same period. Compared with group B, which was injected with phosphate, the number of retinal cells in group A was significantly higher than that in group C , Tissue edema is not obvious; A group of retinal VEGF levels were significantly lower than the B group, and are higher than the C group. Conclusion Angiostatin can significantly reduce the expression of VEGF in retinal tissue of diabetic rats, and can effectively inhibit the changes of retinal edema and internal tissue structure in diabetic rats. Therefore, angiostatin has the potential to prevent and treat diabetic retinopathy.
