目的:优选川党参中核苷类成分的提取工艺条件,为该有效部位的开发提供基础资料。方法:采用超声波提取川党参中核苷类成分,以胞苷、尿苷、鸟苷、腺苷的总提取量为评价指标,在单因素试验基础上,通过正交试验考察不同因素对川党参中核苷类成分提取量的影响。运用HPLC测定胞苷、尿苷、鸟苷、腺苷的含量,流动相甲醇(A)-水(B)梯度洗脱(0~10min,1%~5%A;10~15 min,5%~15%A;15~22 min,15%~17%A;22~26 min,17%~20%A;26~32 min,20%~24%A),检测波长260 nm。结果:各因素对核苷类成分提取量的影响顺序为甲醇体积分数>提取次数>料液比>提取时间,最佳提取工艺为加10倍量水提取2次,每次30 min,川党参中4种核苷类成分总提取量1.523 mg·g~(-1)。结论:建立的HPLC检测川党参中核苷类成分的含量方法准确可靠。优选的提取工艺稳定可行,具有节约成本、快速、高效等优点。
Objective: To optimize the extraction conditions of the nucleosides in Codonopsis pilosula and provide the basic information for the development of this effective site. Methods: The nucleosides of Codonopsis pilosula were extracted by ultrasonic wave, and the total amount of cytidine, uridine, guanosine and adenosine was taken as the evaluation index. On the basis of single factor test, Effects of glycosides on the amount of glycosides extracted. The contents of cytidine, uridine, guanosine and adenosine were determined by HPLC. The mobile phase was eluted with methanol (A) -water (B) gradient (0-10 min, 1-5% A; 15% to 22%, 15% to 17% A, 22 to 26%, 17% to 20% A, 26 to 32%, 20% to 24% A) at a detection wavelength of 260 nm. Results: The order of the influence of each factor on the amount of nucleosides extracted was methanol volume fraction> extraction times> solid-liquid ratio> extraction time, the best extraction process was adding 10 times the amount of water extraction twice, each time 30 min, The total amount of 4 nucleosides extracted was 1.523 mg · g ~ (-1). Conclusion: The established HPLC method for the determination of the content of nucleosides in Codonopsis pilosula is accurate and reliable. The preferred extraction process is stable and feasible, with the advantages of cost saving, rapidity and high efficiency.
