本研究通过建立糖尿病大鼠有氧运动模型 ,探讨有氧运动训练对糖尿病大鼠瘦素 (Lep tin) -下丘脑神经肽Y(NPY)轴的影响。实验选用Wistar雄性成年大鼠 ,体重 195± 10g ,随机分为正常对照组、糖尿病对照组、糖尿病运动组。运动组每天游泳 6 0分钟 ,每周游泳 6天 ,连续游泳训练 6周。记录实验开始及结束时动物体重。测定血糖、血胰岛素浓度、内脏附睾脂肪垫重量、血清Lep tin及下丘脑NPY蛋白浓度和下丘脑弓状核NPY基因表达的非同位素原位分子杂交结果。结果表明 :长期有氧运动使糖尿病大鼠糖代谢紊乱得到改善 ,糖尿病大鼠食欲亢进减弱 ,摄食量减少 ,而内脏脂肪储量上升 ,体重增加。提示Leptin -下丘脑NPY轴可能起着重要的能量代谢调节作用
In this study, aerobic exercise model of diabetic rats was established to investigate the effects of aerobic exercise training on leptin-hypothalamic neuropeptide Y (NPY) axis in diabetic rats. Wistar male adult rats weighing 195 ± 10g were randomly divided into normal control group, diabetic control group and diabetic exercise group. Exercise group swim 60 minutes a day, swimming 6 days a week, continuous swimming training for 6 weeks. Animal weight was recorded at the beginning and at the end of the experiment. The results of non-isotopic in situ hybridization were measured for blood glucose, blood insulin concentration, visceral epididymal fat pad weight, serum Lep tin, hypothalamic NPY protein concentration and hypothalamic arcuate nucleus NPY gene expression. The results showed that long-term aerobic exercise could improve the disorder of glucose metabolism in diabetic rats, reduce the appetite and reduce the food intake, and increase the visceral fat reserves and body weight in diabetic rats. Suggesting that Leptin - hypothalamic NPY axis may play an important energy metabolism regulation
