目的建立测定六味地黄方中有效成分马钱素(loganin)和莫罗苷(morroniside)大鼠血清浓度的固相萃取HPLC法;研究大鼠灌胃给予六味地黄方后血清中马钱素和莫罗苷的浓度变化。方法血清样品经C_(18)固相萃取柱提取后,采用HPLC法测定给药前后大鼠药物血清中马钱素和莫罗苷的浓度,并对该方法进行方法学考察。结果血清中内源性物质不干扰样品中马钱素和莫罗苷的测定,马钱素和莫罗苷的线性关系良好,相关系数分别为0.999 8,0.999 8,方法回收率在95%~105%,提取回收率均大于75%,日内精密度和日间精密度均小于12%。结论该方法具有灵敏、快速的优点,可满足不同配伍六味地黄方有效成分在大鼠体内药代动力学研究的要求。不同配伍对药物血清中有效成分的吸收有一定影响。
Objective To establish a solid phase extraction HPLC method for determination of serum concentrations of loganin and morroniside in Liuwei Dihuang Recipe; To study the effect of intragastric administration of Liu Wei Di Huang Fang on loganin and Mo. The concentration of the glucoside changes. Methods The serum samples were extracted by C 18 solid phase extraction column. The concentration of loganin and moroxyside in the serum of rats before and after administration were measured by HPLC and the methodological investigation was conducted. Results The endogenous substances in the serum did not interfere with the determination of loganin and moroxyside in the samples. The linear relationship between loganin and morroside was good. The correlation coefficient was 0.999 8, 0.999 8. The method recovery rate was 95%. At 105%, the extraction recovery rate was greater than 75%, and intraday precision and daytime precision were both less than 12%. Conclusion This method is sensitive and rapid, and can meet the requirements of the pharmacokinetic study of the active ingredients of Liuweidihuang in different combinations. Different combinations have some influence on the absorption of active ingredients in drug serum.
