目的:研究复方降糖滴丸对实验性糖尿病大鼠血糖、血脂的调节作用及机理。方法采用静脉注射链脲佐菌素,并给以高脂饲料制备实验性糖尿病大鼠模型,以复方降糖滴丸进行治疗,测定模型大鼠的空腹血糖(FBG)、糖化血红蛋白(HbA1c)、血清中胰岛素(Ins)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-c)、低密度脂蛋白(LDL-c)、胰岛素样生长因子-1(IGF-1)、肿瘤坏死因子-α(TNF-α)、肝中的超氧化物歧化酶(SOD)、丙二醛(MDA)、脏器系数(肝、肾、胰)和病理学。结果:与模型组相比,复方降糖滴丸能降低模型大鼠FBg,HbA1c、Ins、T g,TC、LDL-C、TNF-α;升高IGF-1;增加肝组织SOD、MDA。与金芪降糖阳性组相比,复方降糖滴丸多个指标效果优于前者特别是HbA1c、T g,LDL-C(P<0.05)。复方降糖滴丸组能显著降低糖尿病大鼠肝(P<0.05 or<0.01)、肾(P<0.01)脏器系数,复方降糖滴丸给药组能有效改善糖尿病大鼠的胰岛细胞质量和数量,缓解高脂饲料喂养引起的脂肪肝和肾损害。结论:复方降糖滴丸能有效降低实验性糖尿病大鼠FB g,HbA1c,其作用机制主要表现为改善大鼠体内糖代谢和促进胰岛素分泌,增加胰岛素生长因子,改善胰岛素抵抗;复方降糖滴丸能降低实验性糖尿病大鼠的血脂,其可能的机理为通过降低糖尿病大鼠体内的TC、TG,低密度脂蛋白。复方降糖滴丸也能促进肝脏脂质过氧化物代谢,此外尚能提高大鼠体内TNF-α。
Objective: To study the effect and mechanism of Fufang Jiangtang Dripping Pills on blood glucose and blood lipid in experimental diabetic rats. Methods Streptozotocin (STZ) was injected intravenously and the diabetic rats were induced with high-fat diet to treat the model rats with fasting blood glucose (FBG), glycosylated hemoglobin (HbA1c) Serum levels of insulin, triglyceride, total cholesterol, high density lipoprotein, low density lipoprotein (LDL-c), insulin-like growth factor-1 ), Tumor necrosis factor-α (TNF-α), liver superoxide dismutase (SOD), malondialdehyde (MDA), organ coefficient (liver, kidney, pancreas) and pathology. Results: Compared with the model group, Fufang Jiangtang Dripping Pill can reduce FBg, HbA1c, Ins, T g, TC, LDL-C and TNF-α in model rats, increase IGF-1 and increase SOD and MDA in liver tissue. Compared with Jin Qijing hypoglycemic positive group, the effect of compound hypoglycemic dripping pills was better than the former, especially HbA1c, T g and LDL-C (P <0.05). Fufang Jiangtang Dropping Pill group could significantly reduce the hepatic (P <0.05 or <0.01) and kidney (P <0.01) organ coefficient of diabetic rats, and Fufang Jiangtang Dripping Pill group could effectively improve the islet cell mass of diabetic rats And the number of fatty liver and kidney damage caused by feeding high fat diet. Conclusion: Compound hypoglycemic dripping pills can effectively reduce FB g and HbA1c in experimental diabetic rats. Its mechanism is mainly to improve glucose metabolism, promote insulin secretion, increase insulin growth factor and improve insulin resistance in rats. Compound hypoglycemic drops Pill can reduce the blood lipid in experimental diabetic rats. The possible mechanism is to reduce TC, TG and LDL in diabetic rats. Compound hypoglycemic dripping pills can also promote liver lipid peroxide metabolism, in addition can still improve the body of rats TNF-α.
