目的探讨三氯乙烯对大鼠睾丸细胞凋亡、c-myc和p53表达的影响。方法选用雄性SD大鼠15只,随机分为3组,每组5只,实验组用1500和3000mg/kg的三氯乙烯经口灌胃染毒,对照组给予等容量花生油。流式细胞术检测细胞凋亡和细胞增殖,用RT-PCR检测c-myc和p53表达。结果 1500和3000mg/kg组大鼠睾丸细胞凋亡率分别为22.11%和25.96%,明显高于对照组(P<0.01);增殖指数分别为15.51±0.89和13.91±1.02,明显低于对照组(P<0.05),睾丸细胞c-mycmRNA分别为0.698±0.067和0.837±0.062,p53mRNA分别为0.619±0.109和0.761±0.094,均较对照组明显增高(P<0.01)。结论三氯乙烯能明显诱导大鼠睾丸细胞凋亡并抑制增殖,并使睾丸细胞c-myc和p53的表达增强。
Objective To investigate the effects of trichlorethylene on testicular cell apoptosis, expression of c-myc and p53 in rats. Methods Fifteen male Sprague-Dawley rats were randomly divided into 3 groups with 5 rats in each group. The rats in the experimental group were orally administered with trichlorethylene at doses of 1500 and 3000 mg / kg, while those in the control group were given peanut oil at the same volume. Flow cytometry was used to detect apoptosis and cell proliferation. The expression of c-myc and p53 was detected by RT-PCR. Results The apoptosis rate of testicular cells in 1500 and 3000 mg / kg groups was 22.11% and 25.96%, respectively, which was significantly higher than that in control group (P <0.01). The proliferation index was 15.51 ± 0.89 and 13.91 ± 1.02 respectively, which was significantly lower than that in control group (P <0.05). The levels of c-myc mRNA in testis cells were 0.698 ± 0.067 and 0.837 ± 0.062, respectively, which were significantly higher than those in control group (0.619 ± 0.109 and 0.761 ± 0.094, P <0.01). Conclusion Trichlorethylene can obviously induce testicular cell apoptosis and inhibit proliferation in rats, and enhance the expression of c-myc and p53 in testicular cells.
