目的:利用叶绿体matK基因序列探讨重楼属植物的10个样品系统发育及分子鉴定,为重楼的保护和合理利用提基本供理论依据。方法:从分布于湖南省、云南省和吉林省的10个重要的重楼类群样品中提取总DNA,经扩增纯化后,利用PCR产物直接测序。结果:测定了10个重要的重楼类群样品的matK基因序列,序列长度为1 039 bp,比较了该基因的差异,构建了基于matK基因序列的遗传进化树。结论:这10个重楼属植物样品可以划分为4个组群,matK基因序列的结果不支持长药隔重楼、滇重楼、短梗重楼、七叶一枝花成为独立的变种,建议作为多叶重楼的变型处理。
OBJECTIVE: To investigate the phylogeny and molecular characterization of 10 samples of Rheum from the matK gene sequence of chloroplast, and provide the basis for the protection and rational utilization of the re-building. Methods: Total DNA was extracted from 10 important re-arboreal populations distributed in Hunan, Yunnan and Jilin provinces. After amplification and purification, PCR products were directly sequenced. Results: The matK gene sequences of 10 important re-arboreal populations were sequenced. The length of the matK gene was 1 039 bp. The genetic differences were compared and a genetic maturation tree based on the matK gene sequence was constructed. Conclusion: The 10 samples of plants belonging to the genus Torreya genus can be divided into four groups. The matK gene sequence does not support the results of matK gene sequence. As a multi-leaf heavy floor variant processing.
