目的:观察牵牛子水煎液及拆分组分对大鼠阿霉素肾病药效学研究,找寻牵牛子对肾病模型作用的物质基础。方法:采用尾静脉注射阿霉素的方法建立大鼠阿霉素肾病模型。考察牵牛子水煎液及拆分组分对阿霉素肾病模型大鼠血清生化指标及IL-1β和TNF-α的影响。结果:与空白组比较,模型组模型组白蛋白、总蛋白含量明显降低;总胆固醇、总甘油三酯、血肌酐和尿素氮的含量明显升高;IL-1β、TNF-α和尿中蛋白含量明显升高;肾小球上皮细胞肿胀明显,肾小球毛细血管淤血并部分闭塞,系膜基质重度增生呈弥漫性分布。与模型组比较,水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg可以升高白蛋白、总蛋白、血肌酐、尿素氮含量;可以降低总甘油三酯、总胆固醇含量;可以降低IL-1β和TNF-α的浓度;可以降低尿中蛋白浓度。光镜检查结果显示牵牛子水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg能减轻上皮细胞肿胀及肾小球系膜基质增生,改善大鼠肾脏的病理变化。结论:牵牛子水煎液0.63g/kg、多糖0.37g/kg和酚酸0.13g/kg可以提高肾小球的滤过功能,调节血浆胶体渗透压的平衡及血脂紊乱,从而改善肾病综合征表症。作用机制可能通过降低一系列炎性细胞因子的含量,从而达到缓解水肿的疗效。

The results of light microscopy showed that 0.63 g / kg, 0.37 g / kg polysaccharide and 0.13 g / kg phenolic acid could alleviate epithelial cell swelling and glomerular mesangial matrix hyperplasia, and improve pathological changes of rat kidney.
