目的:探讨重楼总皂苷诱导人肺腺癌A549细胞凋亡的作用及对Caspase3、Bcl-2蛋白表达的影响。方法:CCK-8法检测重楼总皂苷对细胞增殖的抑制作用;Hoechst染色后激光共聚焦显微镜观察细胞凋亡形态的变化;Annexin V-FITC/PI双标记流式细胞术检测重楼总皂苷对细胞凋亡的影响;Western blotting检测重楼总皂苷对细胞凋亡相关蛋白Caspase3、Bcl-2表达的影响。结果:重楼总皂苷对A549细胞的生长有显著抑制作用,可引起细胞形态发生明显改变,诱导细胞发生凋亡,显著上调促凋亡蛋白Caspase3的表达,下调抑制凋亡蛋白Bcl-2的表达,且与浓度成依赖关系。结论:重楼总皂苷可显著诱导A549细胞凋亡,其机制与上调促凋亡蛋白Caspase3的表达,下调抑制凋亡蛋白Bcl-2的表达相关。
OBJECTIVE: To investigate the effect of total saponins of Rehmannia glutinosa on the apoptosis of human lung adenocarcinoma A549 cells and its effect on the expression of Caspase3 and Bcl-2 proteins. Methods: The inhibitory effect of total saponins of Reaumuria trigonosa on cell proliferation was detected by CCK-8 method. The morphological changes of apoptotic cells were observed by laser confocal microscopy after Hoechst staining. The total saponins of Reaumuria soongorica were assayed by Annexin V-FITC / PI double-labeled flow cytometry On apoptosis. Western blotting was used to detect the expression of Caspase3 and Bcl-2 on the apoptosis-related proteins. Results: The total saponin of Rhizoma Paridis significantly inhibited the growth of A549 cells, and caused obvious changes of cell morphology, induced cell apoptosis, increased the expression of proapoptotic protein Caspase3 and down-regulated the expression of Bcl-2 , And with the concentration of a dependent relationship. Conclusion: SSP can significantly induce the apoptosis of A549 cells. The mechanism is related to up-regulating the expression of Caspase3 and down-regulating the expression of Bcl-2.
