目的研究温郁金挥发油对β-淀粉样蛋白(Aβ_(25-35))致阿尔茨海默病(AD)模型小鼠的学习记忆改善作用。方法 60只昆明种小鼠随机分为假手术组,模型组,温郁金挥发油低(0.016 mL/kg)、高(0.048 mL/kg)剂量组,盐酸多奈哌齐组(1.30 mg/kg)及参枝苓组(2.6 mL/kg),海马内注射(Aβ_(25-35)),制作小鼠AD模型。连续灌胃给药15 d。Y迷宫和Morris水迷宫实验检测各组小鼠认知行为的改变,HE染色观察其对AD模型小鼠神经元的影响。结果温郁金挥发油可显著增加AD模型小鼠的自发交替反应率,缩短逃避潜伏期,减少总路程,延长游泳路程百分比,具有统计学意义(P<0.05或P<0.01);可改善海马区神经元细胞排列紊乱、水肿等现象,减少神经元死亡。结论温郁金挥发油对Aβ_(25-35)致阿尔茨海默病模型小鼠学习记忆能力具有改善作用。
Objective To study the effects of volatile oil of Wenyujin on learning and memory in Alzheimer’s disease (AD) model mice induced by β-amyloid (Aβ 25-35). Methods Sixty Kunming mice were randomly divided into three groups: sham group, model group, volatile oil of 0.016 mL / kg, 0.048 mL / kg, donepezil hydrochloride 1.30 mg / kg, Group (2.6 mL / kg) and intra-hippocampal injection (Aβ_ (25-35)). Continuous oral administration of 15 d. Y maze and Morris water maze test to detect the cognitive behavior of mice in each group, HE staining to observe the impact of AD model mice neurons. Results Volatile oil from warm turmeric could significantly increase the rate of spontaneous alternation, shorten the escape latency, decrease the total distance and extend the percentage of swimming distance in AD model mice, with statistical significance (P <0.05 or P <0.01); improve the neuronal cells in hippocampus Arranged disorder, edema and other phenomena, reduce neuronal death. Conclusion Volatile oil from Wenyujin can ameliorate the learning and memory abilities of mice with Alzheimer’s disease induced by Aβ_ (25-35).
