对黄芩愈伤组织进行不同聚乙二醇浓度PEG-6000处理培养,探讨不同聚乙二醇(PEG)浓度对黄芩愈伤组织生长及黄芩苷含量的影响.采用不同浓度(0,5%,10%,15%,20%)的PEG-6000模拟水分对黄芩愈伤组织进行处理,测定其鲜重、干重,HPLC法测定黄芩苷含量,计算黄芩苷积累量.结果表明:PEG对黄芩愈伤组织鲜重、干重及黄芩苷含量均有显著影响.在培养周期内随着PEG浓度的升高,黄芩愈伤组织的生长受到不同程度的抑制,且浓度越高,抑制作用越明显,但当浓度为20%时,抑制效果减弱;干重表现与鲜重相反,浓度越高,干重越大,20%处理下的干重最大.黄芩苷含量和积累量在培养周期内随PEG浓度的升高而降低,对照组明显高于处理组,而20%处理下含量有所回升.PEG处理不利于黄芩愈伤组织的生长和黄芩苷的合成,但可以增加干物质的积累,20%处理下黄芩苷含量有所回升.“,”The effects of different PEG-6000 concentrations on callus growth and baicalin content of S.baicalensis were studied. The calli of S. baicalensis were treated with different concentrations( 0, 5%, 10%, 15% and 20%) of PEG-6000. The fresh weight and dry weight were determined, and the content of baicalin was determined by HPLC. The accumulation of baicalin was calculated. PEG had significant effects on callus fresh weight, dry weight and baicalin content. In the culture period, with the increase of PEG concentration, the callus growth of S. baicalensis was inhibited in different degrees, and the higher the concentration, the more obvious the inhibition. But when the concentration was 20%, the inhibition effect was weakened; On the contrary, the higher the concentration, the greater the dry weight, 20% of the dry weight under the largest treatment. The content and accumulation of baicalin decreased with the increase of PEG concentration in the culture period, and ck wassignificantly higher than that in the treatment group, and the content of baicalin was increased. PEG treatment is not conducive to the growth of calli of S. baicalensis and the synthesis of baicalin, but it can increase the accumulation of dry matter and the content of baicalin in 20% treatment.
