采用不同浓度尿素和盐酸胍从小牛骨基质中分离纯化出牛骨形成蛋白(bBMP) ,将其植入小鼠股部的肌肉内观察诱导成骨过程。组织学和酶组织化学研究表明 ,bBMP诱导间充质细胞逐渐分化形成软骨细胞和成骨细胞 ,最终新骨形成。诱导成骨的时间和过程与以往报道一致。bBMP诱导骨形成过程呈现连续又具显著的阶段性。本研究结合组织学和酶组织化学观察的特点 ,将其全过程分为4个时期 ,(1)间充质细胞增生期 ;(2)软骨细胞分化期 ;(3)新骨组织形成期 ;(4)骨组织成熟期。
Bovine bone morphogenetic protein (bBMP) was isolated and purified from the bovine bone matrix using different concentrations of urea and guanidine hydrochloride. The bBMP was implanted into the muscle of the femoral part of the mouse to induce osteogenesis. Histological and enzymatic histochemical studies show that bBMP induces the gradual differentiation of mesenchymal cells into chondrocytes and osteoblasts, eventually resulting in new bone formation. The time and process of induced osteogenesis are consistent with previous reports. bBMP induced bone formation process showed a continuous and significant stage. In this study, histological and enzymatic histochemical observations combined with the characteristics of the whole process is divided into four periods, (1) mesenchymal cell hyperplasia; (2) chondrocyte differentiation; (3) the formation of new bone tissue; (4) bone tissue maturation.
