目的探讨干扰素(Interferon,IFN)及白细胞介素-6(Interleukin-6,IL-6)、IL-1β在肠道病毒71型(Enterovirus 71,EV71)感染过程中的作用。方法采用EV71-FY23,在IFNα1b、IFNλ1和IFNγ分别存在的条件下感染Vero细胞及KMB17细胞,检测IFN对细胞的保护作用;建立EV71感染乳鼠模型,检测IFNα1b、IFNλ1、IFNγ、IL-6及IL-1β的抗病毒作用。结果IFNα1b和IFNλ1对EV71感染的细胞具有明显的保护作用,而IFNγ的保护作用不明显;IFNα1b对感染EV71的乳鼠具有较好的保护效果,其在病毒感染之前的预处理方式产生的保护效果较为明显;IL-1β对感染EV71的乳鼠亦有不同程度的保护作用,而IL-6预处理方式不但无保护作用,反而加速了乳鼠的死亡。结论 IFN和IL-1β对EV71感染具有保护作用,而IL-6无此作用。
Objective To investigate the role of Interferon (IFN) and Interleukin-6 (IL-6) and IL-1β in the infection of Enterovirus 71 (EV71). Methods EV71-FY23 was used to infect Vero cells and KMB17 cells in the presence of IFNα1b, IFNλ1 and IFNγ respectively to detect the protective effect of IFN on the cells. The EV71-infected neonatal rat model was established to detect the expression of IFNα1b, IFNλ1, IFNγ, IL-6 and IFN- IL-1β antiviral effect. Results IFNα1b and IFNλ1 had significant protective effect on EV71 infected cells, while IFNγ had no protective effect. IFNα1b had a good protective effect on EV71-infected neonatal rats, and its protective effect in the pre-treatment of virus infection More obvious; IL-1βinfected with EV71 suckling mice also have different degrees of protection, and IL-6 pretreatment not only without protection, but accelerated the death of suckling mice. Conclusion IFN and IL-1β have a protective effect on EV71 infection, but IL-6 does not have this effect.
