目的通过对乳腺癌细胞系T47D稳定转染,探讨真核表达载体质粒对细胞生物学特性的影响。方法建立稳定转染细胞系,观察细胞形态学改变,检测细胞增殖能力、软琼脂内克隆形成率和细胞迁移能力。结果一株转染细胞系(pcDNA-4)与对照组细胞比较,在形态上发生了明显改变,细胞在软琼脂内的克隆集落形成率显著增高,细胞体外迁移能力显著增强。结论质粒载体DNA稳定转染,可能通过与宿主染色体随机整合,引起多种细胞生物学特性改变。
Objective To investigate the effect of eukaryotic expression vector plasmid on cell biology by stably transfecting breast cancer cell line T47D. Methods Stable transfected cell lines were established. The morphological changes of cells were observed. The proliferation of cells, the rate of colony formation in soft agar and the ability of cell migration were detected. Results Compared with the control group, a pcDNA-4 transfected cell line (pcDNA-4) significantly changed in morphology. The colony formation rate of the cells in soft agar was significantly increased and the ability of cell migration in vitro was significantly enhanced. Conclusion Stable transfection of plasmid vector DNA may cause a variety of cell biological characteristics through the random integration with the host chromosome.
