目的探讨Toll样受体(Toll-like receptor,TLR)2与TLR4在溃疡性结肠炎(ulcerative colitis,UC)SD大鼠结肠黏膜上皮细胞的表达情况,及维生素D3对UC的治疗作用。方法 SD大鼠60只,随机分为正常对照组、UC模型组和维生素D3治疗组,每组20只。UC模型组、维生素D3治疗组采用三硝基苯磺酸法连续灌肠10d,建立UC模型。造模完成后,维生素D3治疗组采用胆维丁乳原液灌胃,按0.005mL/g,1次/d,连用9d;正常对照组和UC模型组大鼠给予生理盐水灌胃9d。各组大鼠均行疾病活动指数(disease activity index,DAI)评分,并对大鼠结肠组织行组织病理学评分(histopathology score,HPS),采用实时荧光定量PCR法检测结肠组织TLR2 mRNA与TLR4 mRNA表达情况。结果 UC模型组DAI评分(8.06±1.95)、HPS评分(14.56±0.84)和结肠组织TLR2mRNA(0.420 00±0.170 00)、TLR4mRNA(1.580 00±1.000 00)表达水平均明显高于正常对照组(0、0、0.000 38±0.000 18、0.000 57±0.000 23)和维生素D3治疗组(5.09±2.16、10.12±2.09、0.027 00±0.019 00、0.018 00±0.015 00)(P<0.05),维生素D3治疗组高于正常对照组(P<0.05)。结论维生素D3能部分抑制TLR2与TLR4表达,对UC有辅助治疗作用。
Objective To investigate the expression of Toll-like receptor (TLR) 2 and TLR4 in colon mucosal epithelium of ulcerative colitis (SD) rats and the therapeutic effect of vitamin D3 on UC. Methods Sixty SD rats were randomly divided into normal control group, UC model group and vitamin D3 treatment group, with 20 rats in each group. UC model group, vitamin D3 treatment group using trinitrobenzene sulfonic acid method of continuous enema 10d, the establishment of UC model. After the model was established, the vitamin D3 treatment group was given gavage solution of gut buttermilk by gavage at 0.005 mL / g once daily for 9 days. Rats in normal control group and UC model group were given gavage for 9 days. The rats in each group were subjected to the disease activity index (DAI) score, and the histopathology score (HPS) of the colon tissues was determined. The real-time quantitative PCR was used to detect the expressions of TLR2 mRNA and TLR4 mRNA Express the situation. Results The DAI score (8.06 ± 1.95), HPS score (14.56 ± 0.84) and TLR2 mRNA (0.420 00 ± 0.170 00) and TLR4 mRNA (1.580 00 ± 1.000 00) in UC model group were significantly higher than those in normal control group , 0,0.000 38 ± 0.000 18,0.000 57 ± 0.000 23) and vitamin D3 treatment group (5.09 ± 2.16,10.12 ± 2.09,0.0200 ± 0.01900,0.018 00 ± 0.01500, respectively) (P <0.05), vitamin D3 treatment Group than in the normal control group (P <0.05). Conclusion Vitamin D3 can partially inhibit the expression of TLR2 and TLR4, and has an adjuvant therapeutic effect on UC.
