[目的]建立人尿中多环芳烃羟基代谢物(OH-PAHs)的超高效液相色谱-串联质谱(UPLC-MS/MS)分析方法,用于同时检测尿中11种OH-PAHs,包括2-羟基萘、1-羟基萘、3-羟基芴、2-羟基芴、2-羟基菲、1-羟基菲、1-羟基芘、3-羟基(蔗)、6-羟基(蔗)、9-羟基苯并[a]芘和3-羟基苯并[a]芘.[方法]尿样经过夜酶解12h、Envi-18固相萃取柱(3mL,500 mg)纯化富集以及氮吹浓缩后,经Symmetry C18色谱柱(250 nn×4.6mm,5μm)分离,采用UPLC-MS/MS(8050)测定.[结果]尿样中11种OH-PAHs在0.002~200μg/L范围内线性关系良好,线性相关系数均大于0.99.加标回收率为71.4%~109.4%,精密度为2.7%~11.6%,检出限为0.001 ~0.040 μg/L.[结论]该方法可同时分析尿中11种OH-PAHs,灵敏度和精密度高,回收率和重复性良好,可用于人群多环芳烃暴露的生物监测.“,”[Objective] To develop an ultrasensitive performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method for simultaneous determination of eleven hydroxylated metabolites of polycyclic aromatic hydrocarbons (OH-PAHs) in human urine,including 2-hydroxynaphthalene,1-hydroxynaphthalene,3-hydoxyfluorene,2-hydoxyfluorene,2-hydroxyphenanthrene,1-hydroxyphenanthrene,1-hydroxypyrene,3-hydroxychrysene,6-hydroxychrysene,9-hydoxybenz [a] pyrene,and 3-hydoxybenz [a]pyrene.[Methods] The urine samples were hydrolyzed overnight for 12h first,then purified and enriched by Envi-18 solid phase extraction cartridges (3 mL,500mg),concentrated by nitrogen blowing,separated by Symmetry C18 column (250mm × 4.6mm,5 μm),and finally determined with UPLC-MS/MS (8050).[Results] The eleven OH-PAHs showed a good linearity in a concentration range of 0.002-200 μg/L and the correlation coefficients were all higher than 0.99.The recovery rates were 71.4%-109.4%,the relative standard deviations were 2.7%-11.6%,and the limits of detection were between 0.001 and 0.040 μg/L.[Conclusion] The established method is applicable to analyze eleven OH-PAHs in urine simultaneously.The method is sensitive and reproducible with high precision and good recoveries,and it is suitable for the biomonitoring of PAHs exposure.
