目的繁育和鉴定Notch 1基因敲除小鼠。方法用B6.Notch 1flox/flox和B6.Alb-Cre转基因小鼠进行饲养和杂交;将繁育出的第1代小鼠再进行相互交配,得到第2代小鼠。通过PCR鉴定出基因型为B6.Notch 1flox/flox.Alb-Cre的第2代小鼠。采用Western blot法检测Notch 1蛋白在肝脏中有无表达。结果两种转基因小鼠繁殖产生的第2代小鼠基因型符合孟德尔遗传定律,可通过该方法获得较多的B6.Notch 1flox/flox.Alb-Cre小鼠。该基因型小鼠肝脏中不表达Notch 1蛋白。结论应用Cre-loxp系统可以成功地敲除小鼠Notch 1基因。
Objective To breed and identify Notch 1 knockout mice. Methods B6.Notch 1 flox / flox and B6.Alb-Cre transgenic mice were bred and crossed. The first generation of mice were bred to each other to obtain the second generation of mice. Generation 2 mice of genotype B6.Notch 1 flox / flox.Alb-Cre were identified by PCR. Western blot was used to detect the presence or absence of Notch 1 protein in the liver. Results The second generation mouse genotypes produced by the two transgenic mice conformed to Mendel’s law of inheritance, and more B6.Notch 1 flox / flox.Alb-Cre mice could be obtained by this method. Notch 1 protein was not expressed in the liver of this genotype mouse. Conclusion The mouse Notch 1 gene can be knocked out successfully using Cre-loxp system.
