分析11-羰基-β-乙酰乳香酸(AKBA)对小鼠黑素瘤细胞系B16F10增殖和凋亡的影响,探讨其作用机制。AKBA是从天然乳香中提取的单体。通过CCK-8法测定AKBA在不同浓度及不同时间对B16F10细胞增殖的影响;以流式分析法检测AKBA对B16F10凋亡的作用;以Real-time PCR法检测细胞增殖及凋亡相关基因的表达;以Western blot法检测细胞增殖与凋亡相关的NF-κB信号通路的变化。结果显示,AKBA可以明显促进B16F10细胞的凋亡,抑制其增殖,并与NF-κB信号转导通路的活性有关。
The effects of 11-carbonyl-β-acetyl boswellic acid (AKBA) on the proliferation and apoptosis of the mouse melanoma cell line B16F10 were analyzed, and the mechanism of action was explored. AKBA is a monomer extracted from natural frankincense. The effect of AKBA on the apoptosis of B16F10 cells was assayed by flow cytometry (FCM), the proliferation of B16F10 cells was assayed by CCK-8 assay, and the proliferation and apoptosis-related genes expression of AKBA were detected by Real-time PCR ; Western blot was used to detect the changes of NF-κB signal pathway in cell proliferation and apoptosis. The results showed that AKBA can significantly promote the apoptosis of B16F10 cells and inhibit its proliferation, which is related to the activity of NF-κB signal transduction pathway.
